All other things being equal the 6" will have better potential accuracy due to the longer sight radius. But the difference is small and accuracy is going to be more dependent on the shooters abilities.

Rover is correct, the cylinder on the Six is slightly bigger, but still designated with the same holster size. I have a 4" 66-1 and a 4" Security Six and they both use the same holster, so I would guess if you find a k-frame holster with the same barrel length it should suffice.
I see the Speed Six as something of interest to the hard core Ruger collector. As a concealed carry gun, the SP101 is better suited. I have a Security Six 357 and a Service SIx 38 Special. The 357 is just an exceptional firearm (with its action job). To me it's the perfect size for packing. I'd use .38s for practice & .357s for carry.The Ruger Security-9 weighs in at 23.7 ounces when empty. This is a generally-acceptable for concealed carry. However, loaded it weighs close to 27 ounces, which is quite a bit heavier and some may feel it.
